The Pilgrims, who were 1-0 victors at the KAL Group Stadium in pre-season, begin their latest adventure in the world's premier cup competition (kick-off 3.00pm).
Trinity recorded home wins over AFC Telford United and Grantham Town – in addition to an away win over higher ranked Darlington – to reach the final qualifying stage.
With £9,375 in prize money – alongside a place in Monday evening's televised first round draw – there is plenty at stake as two of Lincolnshire's non-league sides go head-to-head.
There has not been a competitive non-Lincolnshire Senior Cup meeting between the sides since New Year's Day 2018, when Reece Thompson equalised for the Pilgrims in a 1-1 draw at The Northolme.
Should Saturday's tie end all-square, a replay will take place at the Jakemans Community Stadium on Tuesday (October 15, kick-off 7.45pm).
