The Pilgrims will welcome the EFL Championship club to the Jakemans Community Stadium for their opening Group C fixture, which will see later matches take place against Sunderland, Newcastle United and Middlesbrough.
Should the match remain level after 90 minutes, a penalty shootout will take place to determine the destination of an extra point.
United boss Ian Culverhouse is set to shuffle his selection pack, with the likes of Tom Leak and Adam Marriott among those in contention to start in pursuit of valuable minutes.
A number of Joe Simpson’s crop of under-23s players could be involved against the Whites.
Kick-off at the Jakemans Community Stadium is 7.30pm.
