The Pilgrims, who are set to hand opportunities to a host of emerging talent at the club, will entertain Lincoln City (kick-off 7.45pm).
The likes of Jake Lovelace, Tyler Knowles, Will Hall, Ryan Hand and Sam Essien are poised to feature for the Pilgrims as Joe Simpson leads a United side into knockout action at the Jakemans Community Stadium.
The Pilgrims have not lifted county honours since 2013, when they defeated Gainsborough Trinity in the final of the Lincolnshire Senior Shield.
United and City both received byes to the quarter-final stage as two of the four highest-ranked clubs in the county, alongside Grimsby Town and Scunthorpe United.
Should the tie remain level after 90 minutes, the match will head to penalties.
