The winners will pocket £9,375 in prize money and advanced to Sunday's televised first round draw.
The sides have not met competitively since the late-1960s, with Stratford currently playing in the Southern League Premier Division Central.
The Bards were 3-2 victors over Long Eaton United in the previous round, with United reaching the final qualifying stage courtesy of routine home wins over Corby Town (6-0) and East Thurrock United (4-0).
Manager Craig Elliott expects to have a fully-fit squad to choose from, with full-back Matt Tootle expected to return to contention following illness.
Joe Leesley, who made his long-awaited return to the starting XI on Tuesday evening, will be among those hoping to start against the Warwickshire outfit.
Should a replay be required, it will be staged on Tuesday October 19 (kick-off 7.45pm).
