The Black Cats' under-21s will visit the Jakemans Community Stadium for a second season running - following the Pilgrims' 1-0 win in November 2024.
This time around, a three-point haul for Graham Coughlan's side could lift the Pilgrims to the brink of qualification from Group D, having already defeated Middlesbrough and Leeds United in their opening two fixtures.
Coughlan has indicated that he will shuffle his pack as much as competition rules allow - with the likes of goalkeeper Bobby Jones and strikers Lenell John-Lewis and Kieren Donnelly among those chasing valuable minutes.
Donnelly has netted three goals in two National League Cup encounters so far this season.
Sunderland were 3-2 victors at Scunthorpe United in their opening Group D fixture.
